Research context

Selank, also indexed as TP-7, is a synthetic heptapeptide built from tuftsin, the Thr-Lys-Pro-Arg tetrapeptide derived from the CH2 domain of the immunoglobulin G heavy chain, extended at the C-terminus with Pro-Gly-Pro. It was developed in Russia from structure–activity work on tuftsin analogs, and the same Pro-Gly-Pro design appears in the related peptide Semax.

The extension is the point of the molecule. Tuftsin itself is cleaved rapidly by amino- and carboxypeptidases; appending Pro-Gly-Pro places prolines on both sides of the new C-terminal region, and proline-adjacent bonds resist most exopeptidases. Tritium-labelling studies of Selank and its fragments in rodent tissue and plasma were used to map that degradation pattern and to show that the Pro-Gly-Pro tail is itself a released metabolite with its own reported activity. The peptide carries one lysine and one arginine, so it is basic and highly water-soluble, and it has no disulfide or cyclic constraint.

At the molecular level the reported pharmacology is enzymatic and transcriptional rather than receptor-selective. Work using tritium-labelled Leu-enkephalin showed Selank inhibits enkephalin-degrading enzyme activity in plasma, which is the mechanism most often cited for its effects on opioid-peptide turnover in rodent models. Two studies in Frontiers in Pharmacology (2016 and 2017) reported changes in the expression of genes involved in GABAergic neurotransmission after Selank exposure, in rat tissue and in the IMR-32 neuroblastoma line respectively, and other work describes effects on serotonin metabolism and on monoamine- and neurotrophin-pathway transcripts. Its tuftsin-derived immunomodulatory activity is studied separately.

For a laboratory, Selank is mainly a reference and comparator peptide: an enzyme-inhibition substrate or inhibitor in enkephalinase assays, a stimulus in gene-expression panels on neuronal lines, a comparator in rodent behavioural pharmacology, and an analytical standard for LC-MS confirmation. Its small size, high solubility and lack of oxidation-prone residues make it an easy positive control when validating generic short-peptide chromatography, and the Pro-Gly-Pro tail means degradation studies can follow both the parent heptapeptide and its released fragment in one run. It is also a recurring analyte in forensic and anti-doping chemistry — a 2020 study in Drug Testing and Analysis characterized Selank among the putative cognitive-enhancing peptides identified by LC-MS in seized preparations, which makes a well-characterized reference lot useful for method development and confirmation work.

Regulatory position, stated factually: Selank is registered as a medicine in Russia and is not FDA-approved in the United States. FDA lists selank acetate (TP-7) among bulk drug substances whose compounding nominations were withdrawn by the nominators, and Selank was not among the peptide substances covered by the October 2026 HHS interim enforcement-discretion notice.
